Yes it will. You will only need about a 1/4 inch layer of casing placed on top of the substrate at around 90% colonization.

Q: How long can this be stored at room temp?

If stored in the original packaging the casing can keep around 3 months.

Q: Am I able to mix mycilum to the bag without compromise?

Yes just keep it clean. You can mix a jar of mycilium after you break it up real good with gloves on then open the bag and pour it in shake it up and put her in the dark. Fruits will happen soon. I like this company.

One bag will be enough. You will only need to apply a 1/4 inch think layer.
