Guttermann thread is 50/3 which means it's three strand 50 wt finer thread. The bigger the number the finer the thread and being 3 ply is very strong. Do I need to use a thread holder for this large spool of thread?

No, the spool is not that big where you have to use a thread holder. It fits fine on my regular spool holder on my machine.

How do i know if this is tex 50 or 70 and what size needle?

The Gutermann Sew-All thread is usually tex 30. The needle size recommended range is #10 - 14.
