Do you have to use the grease gun to use this product? I don't have one, and was just hoping to scoop out the grease to pack my mixer's gears.

It comes in a tube that you can insert into a grease gun but you can do what I did and just pop the black plastic cap off and scoop out the right amount of grease.  Take a photo of the gears before you start cleaning out the old grease so you have a better idea of putting in the right amount.  Cleaning out the old grease is a little tedious, but it is important to get the old grease out as once it starts to liquefy and leak, it will just ruin the new grease if you don't get it all out.  Qtips are helpful for the final cleanup.  After you have it back together the mixer will sound a little different at first, but after the first couple of batches of cookies or whatever you are making it will smooth right out. Does it fit the speedplay grease gun?

Yes but order the small size tube the two or three ounce one not the large one.   I use this grease with the Speedplay gun.  Very effective

Is it Lithium Based?

No it's not Lithium based, it's basically SYNTHETIC OIL based, has ( PTFE ) in it short for POLYTETRAFLOUROETHYLENE. They also call it SYNCOLON, claims to be one of the most slippery surfaces in the world ? Remember the non-stick frying pans ? They were coated with PTFE basically Teflon ? I use it on EVERYTHING EVEN MY GUNS, WHEEL BEARINGS, GREAT STUFF, FOOD SAFE, VERY VERY STICKY.

I called the Synco Chemical Corp, 631 567 5300 and was told that the lube you are inquiring about was manufactured on 2/17/2015.   15048 B27 is read as the 15th year and the 027 day of the year, therefore, 2/17/2015.  B27 stands for the 27th batch.    This lubricant has unlimited use of time as long as it's in its original container.
