Would this work for pressing two pieces of paper that are glued together with a Crayola glue stick to help eliminate wrinkles?

I think it could work in that capacity At least I’m sure it would help to squeeze out all the excess glue which would help flatten the piece.

Can this be used with fabric paint?

Not sure, I use it to lay my vinyl on my cricut mat.

Can you tell the measured durometer of this roller? (Speedball deluxe soft rubber brayer on Amazon indicates 40/42). How does this compare?

I would say hard rubber... when I press down and roll on the "drills"(I call then diamonds) there is no imprint left in roller.... seems very durable .... easy to clean although I have not tried to remove the roller for handle
