Can I use this with an acrylic-lacquer spray paint (automotive touch-up)?

Yes just make sure it's completely dry before applying top coat

Can this be used on a previously spray painted surface (scuffed with 80 grit) or is a bare surface recommended?

It would stick but IMO the surface would not be very smooth. This will fill some small areas If this is what you want other wise I would strip the piece and then apply the primer. The better the base coat the better the coverage.
