Certainly hard enough. Not sure how to respond to waterproof. It can probably take some light rain, but with a zipper, I would be suspicious that it is fully waterproof.

Will this hold two 5 tb seagate backup plus drives?

yes but one side of it is zippered the other isnt but i got mine holding 2 drives and 3 or 4 cables total in it its pretty decent size about 3 inches thick in the middle give or take the foam padding. I would say it would fit 2 drives if they are of the 2.5 size variant
